A STUDY TO ANCHOR HCG ON POLYSTYRENE FOR IMMUNOANALYSIS OF BETA-HCG

Abstract

Measurements of beta form of human chorionic gonadotropin (hCG) provide important information in a variety of clinical situations and prenatal screening. In this study, we optimized conditions to absorb hCG antibody onto polystyrene (PS) substrates for immunofluorescence analysis of hCG. The conditions were monitored by evaluating the photoluminescent (PL) intensity obtained upon using Eu(III) labelled hCG antibody and PL enhancement solution. The optimized concentration of hCG antibody in the absorbent solution was found to be 2.5μg/L and the absorption time was 24 hours. The reactivity of the absorbed hCG antibody decreased negligibly after three weeks of storing at 4oC. The immunofluorescence procedure based on the hCG/PS substrates was highly selective to beta-hCG, giving good analytical results when compared with standard Kit. Lower limit of detection (LOD) and lower limit of quantification (LOQ) of the analytical procedure were estimated to be 11.9 and 17.9 ng/ml, respectively. The results demonstrated in this paper provide a cost-effective and precise method for quantification of beta-hCG based on hCG antibody absorbed on PS substrates.
