Immune response stimulated by recombinant PMT vaccine against Pasteurellosis caused by Pasteurella multocida in pig

Abstract

In this study, we assessed the ability in creating immune response in pig, stimulated by R-PasVAC vaccine against Pasteurellosis from PMT based antigens, including Tox1 and tPMT-C780. The purified antigens were mixed with Montanide ISA 201 adjuvant to make the vaccine in which the final concentration of each antigen was 250µg/ml and the adjuvant concentration was 50% v/v. The results of vaccine efficacy assessment showed that the
vaccine strongly stimulated the specific humoral immune response against PMT demonecrotic toxin and protected well mice and pigs against P. multocida bacteria. The optimized doses of vaccine that were determined to be 0.1ml per mouse by subcutaneous route, twice at 2 week interval, and 1ml per 3-4 week old pig by intramuscular route without a booster dose, exhibited
high protection efficacy up to 100% on mice and 90% on pigs. These studied results are very important for further developing R-PasVAC vaccine against Pasteurellosis in pigs based on PMT antigens of Pasteurella multocida strain isolated in Vietna
