Assessment of biological characteristics and effect of Bully dog semen preservation in extender adding ascorbic acid and chicken egg yolk at 5oC

Abstract

The objective of this study aimed at determining the influence of adding ascorbic acid and chicken egg yolk in TCG extender at 5oC for improvement of semen preservation effect of Bully dog breed raising in Viet Nam. The contents of chicken egg yolk and ascorbic acid adding in TCG extender were 0, 5, 10, 15, 20% v/v, and 0, 1, 2, 3% w/v, respectively. Good quality sperm samples were collected from three individual Bully male dogs raising in the Experimental Animal Farm, Viet Nam National University of Agriculture. The preserved semen quality parameters including sperm activity (ratio of progressive sperms) and the rate of abnormal sperm (%) were evaluated at four preserved durations: 0, 24, 48 and 72 hours.The studied result showed that semen extenders adding 2% w/v ascorbic acid or 15% v/v chicken egg yolk, at 5oC had given the motility of sperm increasing 10% higher than that of the control ones and the extenders adding other ascorbic acid or chicken egg yolk contents, also decreasing the abnormal sperms during period of conservation. The result also showed that simultaneous addition of 2% w/v ascorbic acid and 15% v/v chicken egg yolk in extender had increased the sperm motilities at least 10% more and decreased the abnormal sperms apparently.
