RESEARCH THE EFFECT OF PLANT GROWTH REGULATORS ON MORPHOGENESIS FROM DIFFERENT EXPLANT SOURCES OF Eurycoma longifolia

Abstract

This study was conducted to investigate the effect of plant growth regulators and the type of explants on morphogenesis in Eurycoma longifolia. The results showed that, for the cotyledon explants: the rate of callus formation and the fresh weight were the highest, 96.3% and 258 mg respectively when the explants were cultured on the MS medium supplemented with 3.0 mg/L NAA in combination with 0.5 mg/L BA, the calli were yellow and friable; for young leaf explants: the rate of callus formation and fresh weight reached the highest, 100% and 99 mg respectively when the explants were cultured on the MS medium supplemented with 0.5 mg/L NAA in combination with 0.5 mg/L BA, the calli were white and friable; for young petiole explants, the rate of callus formation and fresh weight were the highest 100% and 56 mg respectively when the explants were cultured on the MS medium supplemented with 1.0 mg/L NAA in combination with 2.0 mg/L BA, calli were white and friable. The callogenesis time of the petiole explants occurred earlier than callogenesis time of leaf and cotyledons explants. The callus formation rate of treatment using BA combined with TDZ was lower than treatment using BA combined with NAA. The results of this study show that the collected calli have different morphology and colors. This will be a diverse source of materials to serve many different research directions such as embryogenesis, cell suspension culture to receive secondary compounds... In the study of adventious rooting of in vitro leaf explants, NAA is suitable for the formation of adventious roots of Eurycoma longifolia. The research results showed that the average rooting ratio, number of roots/explants and average root length reached the highest, 80%, 2.5 roots/explants, 23.8 mm respectively in the treatment using 1.0 mg/L NAA. There was no root formation in ex vitro leaves explants in all treatments.
