CHARACTERIZATION OF atpB-rbcL AND trnL DNA SEQUENCES OF Hoya lockii

Abstract

Hoya lockii  is endemic to Viet Nam and currently endangered. Hoya lockii is one of the plants used as ornamental plants as well as medicinal plants. The classification of plants is usually based on morphological characteristics and DNA barcodings such as matK, ITS, rpoC1,... Molecular characteristics are necessary for studying the molecular taxonomy of each species, Therefore, additional molecular sequences are needed to find the most specific markers for each species. In this study, the gene sequences atpB-rbcL and trnL in chloroplasts were isolated from samples of Hoya lockii . The obtained atpB-rbcL gene sequence has a size of 713 nucleotides and the trnL gene has a size of 524 nucleotides. Sequences with high similarity when analyzed by Blast with sequences of the same genus have been published. Both amplified DNA of the two regions rbcL and trnL were used to construct the phylogenetic tree.
