Analysis and determination of protein components of Fasciola gigantica by Western Blot method

Abstract

Based on morphological characteristics and body size indexes of Fasciola spp., two bovine liver fluke species circulating in cattle and buffalo in Bac Giang province, Viet Nam were identified, such as Fasciola gigantica and hybrid form of Fasciola sp. In order to detect the protein components with high antigeniCity for serodiagnosis of fasciolosis in cattle and buffalo, the excretory-secretory and somatic antigens of Fasciola gigantica were extracted, analyzed and compared in this study. The result of SDS – PAGE electrophoresis on 15% polyacrymide gel, staining Coomassive blue and silver showed that 26 somatic antigen protein bands of Fasciola gigantica having molecular weight from 10k Da to 130 kDA and 15 excretory-secretory antigen protein bands having molecular weight from 10 kDa to 70 kDa were identified. By applying the Wetern Blot method, the antigen-antibody reaction in Fasciola infected cattle serum was detected on PVDF membrane. The excretory-secretory antigen protein bands reacted with sero-antibody having molecular weight of: 24, 27, 36; 38, 45, 55, 70, 102 kDa respectively and the somatic antigen protein bands reacted with sero-antibody having molecular weight of 21, 24, 27, 38, 70 kDa respectively.
