Study on In vitro Propagation of Miltonia sp.

Abstract

     In the present study, 3 - 4 protocorms derived from meristem of red and white-pink Miltonia sp. were used as material source for all in vitro culture experiments. Effect of MS medium concentration, coconut water and BA alone or in combination with αNAA on protocorm formation and shoot growth were studied. After 90 days of culture, the results showed that half MS medium supplemented with 15% coconut water, 1.5 mg/l BA in combination with αNAA of 0.1 mg/l, 30 g/l sucrose, 8 g/l agar, 1 g/l activated charcoal, and pH 5.8 was the best for protocorm induction and

shoot growth (25.60 shoots/explant, height of 1.95cm). Half MS medium containing 0.1 - 1 mg/l αNAA or 0.1 - 1 mg/l IBA, 30 g/l sucrose, 8 g/l agar, 1 g/l activated charcoal, and pH 5.8 was suitable for in vitro root formation after 40 days of culture and the rate of shoot formation was 100%. Coconut fiber and fern fiber were suitable substrates for plantlet growth in the greenhouse, with coconut fiber being better in terms of number of roots/explant, root length and survival rate comapred to fern fiber substrate.
