POLYMORPHISM OF DEHYDRIN GENES OF SEVERAL VIETNAMESE MAIZE CULTIVARS

Abstract

Dehydrin - LEA (late embryogenesis abundant) - Dl1 is a plant protein family responded to stress such as drought, low temperature, high salt. They are found in almost plant species. In order to study on genetic diversity of maize, dehydrin genes were amplified by PCR using total genomic DNA of maize cultivars with different drought ability: VN5 is a sticky maize cultivar, Q411- local Hagiang cultivar, HQ2000 - cultivar with high quality protein and LCH9 - high yield cultivar. PCR products were cloned and sequenced. The genes from VN5, Q411 and LCH9 have 628 bp in length, including two exons of 507 bp and an introns of 121 bp: while the gene from HQ2000 have 631 bp in length, including two exons of 507 bps and an intron of 124 bp. The coding sequences have similarity of 98,4 - 99,6%. The putative amino acid sequences have 168 amino acid in length with YSK2 structure and have changed significantly polymorphism. These genes were registered in EMBL database with Accession numbers AM495893, AM495892, AM495891 and AM495894 for cuItivars VN5, Q411, HQ2000 and LCH9, respectively.
