REGENERATION AND STORAGE OF PHALAENOPIS ARTIFICIAL SEEDS

Abstract

Gladiolus propagation using ensuring in vitro conn formation is a high rate of induced corms that servive and development in the greenhouse. Moreover, these conns are more appropriate to preserve and transport than plantlets in order to  ensure that seedling source is delivered to farmers timely and sufficiently. In this study, the effects of sucrose. IBA concentrations, and-culture conditions such as light intensity and culture vessels were studied. The results indicated that high concentrations of surcose (60 - 100 grl) and 0,375 mgr1 IBA in 112 MS medium induced the fonnation of conns from bright red Gladiolus shoots and increased the quality of those corms as welL Besides, shoots cultured in 500 ml vessels and illuminated by 3000 lux light source formed more and better corms, which survived and developed well when transferred to ex vitro condition. Corms weighing more than 0.35 g form plantlets to 100%, while the others also have a high rate of plant establishment (about 90%). This advance is being applied successfully in Dalat Affiliate Institute of Biology, and could be disseminated to help producing a great amount of in vitro corms with high quality and survival rate in order to supply the materials for growers.
