Establishment of the Protocol for Hong Hoang (Cymbidium iridioides) Propagation by Tissuse Culture Technique

Abstract

The propagation of the endangered species Cymbidium iridioides (Hong Hoang) by tissue culture is an urgent requirement in Viet Nam for commercial production. The aim of this study was to establish a successful protocol for in vitro propagation of that species. The experiment was arranged in a randomized complete block (RCB) with 3 replications. Using 4 - 6 cm young buds or seed as explants, the best media for initial establishment of the culture were MS + 2% saccharose + 0.65% agar + 1.5 ppm BA, (or 2 ppm Kinetin)/liter for apex regeneration and MS + 1% saccharose + 0.1% peptone + 0.1% active charcoal + 0.56% agar/liter for seed germination. The optimal medium for thin cell layer culture was MS + 1 ppm K + 2% saccharose. Media containing MS + 2% saccharose + 1.0 ppm Kinetin (or  0.5 ppm BA) + 0.65% agar and MS + 0.1% active charcoal  + 2.5%  saccharose were identified to be optimal for  propagation and regeneration, respectively.
