Study on Rapid Micropropagation of Ligusticum wallichii Frach

Abstract

Research was carried out in order to develop a protocol for in vitro rapid propagation of Ligusticum wallichii Frach. using shoot tips. Using 0.15% aqueous mercuric chloride (HgCl2) solution for 10 min appeared to be optimal for surface disinfection of shoot tip explants with survival rate of 68%. MS medium supplemented with 0.75 mg/l BA + 0.1mg/l α.NAA and 10% coconut water or 0.5 mg/l kinetin + 0.1 mg/l α.NAA and 10% coconut water increased the shoot multiplication rate (7.3 times). The best media for rooting of the in vitro shoots were MS + 0.1 mg/l IBA + 0.5 g/l activated charcoal or MS + 0.3 mg/l α.NAA + 0.5 g/l activated carbon with 100% of the shoots having roots within 14 days. The most appropriate time for successful transplantation of in vitro plantlets to soil was from 12/1 to 22/2 with the rate of survival approaching 100% and all the plantlets showed good growth and development.
