Study on Micropropagation of Cyperus corymbosus Lam

Abstract

This study was conducted in order to establish a protocol for rapid propagation of Cyperus corymbosus Lam. from dormant-bud stem. The results indicated that suitable sterilization time of Cyperus corymbosus Lam. with HgCl2 0.1% was 13 minutes. MS medium containing 1.5 ppm BA and
30 g/l sucrose was found to be the most suitable for in vitro shoot regeneration, the rate of regenerated shoots was 1,86 shoots/explant after 4 weeks. The highest rate of shoot propagation was 2.24 times/4 weeks, the average height of shoot was 2,86 cm on MS medium adding 1.5 ppm BA and 0,25 ppm IBA. Addition of 1g/l activated charcoal was positive for root formation with 5,98 roots/shoot and the average height of root was 3.77 cm. Liquid shaking culture was proved to be most suitable for shoot multiplication and the rate of shoot propagation was the highest among all other culture methods including agar medium, agar - liquid medium, semiagar medium, liquid medium with 2.81 times after 4 weeks. Well - rooted plantlets were successfully transplanted to mud-based substrate, the rate of plant survival was 100% and the plantlets developed well.
